A Hollywood, durant la nuit, Rita, une jeune femme, devient amnésique suite à un accident de voiture sur la route de Mulholland Drive. Elle fait la rencontre de Betty Elms, une actrice en devenir qui vient juste de débarquer à Los Angeles. Aidée par celle-ci, Rita tente de retrouver la mémoire ainsi que son identité. Avec Lost Highway et Mulholland Drive, David Lynch s'impose comme l'un des cinéastes les plus étranges et les plus fascinants du moment, malgré les difficultés de compréhension que suscitent généralement de son œuvre. Abandon des règles du cinéma traditionnel, refus de la médiation des idées et de l'intellectualisation de son art par le réalisateur, tels sont ses principes. L'énergie qu'il insuffle au récit, l'étrangeté des personnages, la singularité des situations, la recherche visuelle, toute cette mosaïque démente, seul un artiste de la dimension de David Lynch pouvait l'imaginer et la faire exister. Along Mulholland Drive nothing is what it seems. In the unreal universe of Los Angeles, the city bares its schizophrenic nature, an uneasy blend of innocence and corruption, love and loneliness, beauty and depravity. A woman is left with amnesia following a car accident. An aspiring young actress finds her staying in her aunt's home. The puzzle begins to unfold, propelling us through a mysterious labyrinth of sensual experiences until we arrive at the intersection of dreams and nightmares.

Nach einem Autounfall auf dem Mulholland Drive taumelt die einzige Überlebende, eine benommene Schwarzhaarige durch Los Angeles und findet schließlich Unterschlupf in einem kurzzeitig unbewohnten Appartment, in das bereits am nächsten Tag die herzensgute Betty, eine junge Frau die in Hollywood Kariere als Schauspielerin machen möchte, einzieht. Gemeinsam versuchen sie die Identität der gehemnissvollen Schwarzhaarigen, die sich seit ihrem Unfall an nichts mehr erinnern kann, herauszufinden.
